Ascites fluid is one of the most cost-effective methods in generating antibody from hybridoma cell line. At GeneMay, we use primed adult Balb/c or any other requested strain of mouse for ascites production. For untested cell lines GeneMay can produce ascites for you even if you don't know or can't prove the serological status of your cell line.
After the injection of the hybridoma cell line, animal is monitored daily. Once the ascites fluid is produced,it is harvested by gentle aspiration of the abdominal cavity of the mice. With each successive tap to collect the ascites fluid, the quantity of ascites fluid will decrease, while the antibody concentration increases. The last ascites collection contains most of your antibody . The pooled ascites are then clarified by centrifugation and gauze filtration. Average ascites volume is 3-5 mg per ml. A typical production run takes approximately 2 to 4 weeks to complete.
